Applications and Impact of CRISPR/CAS9 in Bioprocessing
"Today. I'm joined by fanling. Wong director of cell line development and protein sciences and zane starkey wolf director of corporate development from wishy biologics. I'm excited to be speaking with both of you. Today about crisper cast nine technology and its possibilities in the discovery and development biopharmaceuticals. We will also conduct a deep dive on its potential impact on bioprocessing and bio manufacturing. Welcome fanling and zane to the podcast. Zinke randy glad to be here. Thank you brandy before we get too far and because of our audience is quite diverse with regards to their experiences in life sciences. Fenland could you please provide some background on. Crisper cast nine as molecular biology. Gene editing tool. Yeah sure so christmas. Nineteen action system is actually adapted from a natural procure arctic defense mechanism to bacteria to simplify. What could spec assistant can do is took leave the face she and i was. It has been incorporated into the bacteria routine on so that to keep the fate from reproducing. Crisper is actually akron stands for clusters servers regularly interspace. Shot had a dramatic repeats and kissed by the most well well-researched variant of the class outcasts nucleus. Which has been used within the gene editing function. So i think the research community have actually adapted this mechanism to revolutionize how we perform the genetic modifications Not only in pro arctic. But or so. You can arctic sales since the system was first published and zane i know from our previous conversations that you were saying that crisper casts has an intriguing origin. Would you be able to elaborate. Yes interesting research can be found on crisper that dates back to the late. Eighty s Other work has been conducted throughout the first decade of the two thousand however it wasn't until two thousand twelve that two pivotal research papers were published in the journal. Science one by jennifer down nov uc berkeley and manual shopping chair of the university of vienna and then another pianist by doctors cross unanimous and sickness at vilnius university. All demonstrating the use of bacterial. Crisper cast nine as simple programmable. Gene editing jewel. But i know that the story doesn't stop there does it no. It certainly does not in less than a year in two thousand thirteen. The labs of dr fung jong and will chong of the burden student. Mit dr george. Church's lab at harvard reported success in adapting. Crisper cast nine for genome editing in your area cells and both mouse and human cells. And i know that we could really spend an entire podcast. Just on the history of crisper so i wanted to stay focused on the technology here. There's been a lot of excitement since discovery about this molecular biology tool. Can you explain why. Sure the remarkable functionality of this tool is that it allows scientists to target specific locations within the genetic code of an organism to cut out or replace a segment of dna due to the high specificity and exactness of utility. The applications have far reaching potential. And it has already become a much to walks game changer. In many fields of life science because it enables efficient cost effective and precision gene. Editing that has a wide utility for development of biological therapeutics including so and gene therapy disease modeling diagnostics agriculture industrial biology and more. And this has me thinking just about alternatives to crisper casts altogether Are there other ways to edit genomes. And if so what makes a crisper cast so much better. Great question brandy many of the other gene editing systems utilize today such as zinc finger. Nicholas's talons the use of mega nick. Liaises or other by all vectors like a. Iv compared with christopher cast nine are in the end very complex and time intensive often requiring many more steps and thus are more costly as well also and this may be greatest benefit. Is that crisper chess. Nine as a low off target affects profile which again makes it an ideal gene. Editing tool justify along with that. I've read many Recent advances using crisper technologies. Could you elaborate a little bit on those. The advances are extensive and continuous. We speak one example includes crisper a crisper i which are techniques to up and down regulate gene expression using dead cast nine dead cast nine removed the nucleus capability of cast nine but still allows for the targeted binding to a double stranded. Dna sequence of interest using the highway. Specific guide are a that is one of the cornerstones of crisper genome editing. I'd like to add that another application. It's a using crisper for hamas directed repel or so called a the are so this technique in simple terms can repel a double stranded. Dna break which is very important for genus ability. But what does the crisper made. Sdr can do is that. It cannot only to repel a break. But or so crew. Eight the break and then replace it with a small mutation or as elijah sequences so this techniques have actually substantially opened ability or researchers to make gino added more quickly and more efficiently

ECB announces more stimulus and less growth
"Now, starting with an ECB preview central banks takes takes center stage, this morning that descends upon Vilnius Lithuania for his rate decision today's Caroline, just said Bloomberg's peo- skill. Most ski has the preview. The expected to offer generous terms on loans to banks when policymakers. And now they're this after the meeting in Vilnius prison. Mario drug Ethan, like escape questions about slowing inflation and the impact that is going to think pensions could have on the year area Konami with any cutting rate and opening the possibility to follow suit. The could find himself under pressure to deliver more stimulus down the line in Frankfurt gives cutting Muskie, Bloomberg daybreak Europe. So let's just say well Cranfield for my markets. Live team joins us central banks as we say taking center stage, the decision. Will it bring an end to the valley? Very good question. I'm sure a lot of people would like to know about it. Probably depends how the equity market in your decision today. What people are expecting as was mentioned. There is size stimulus of mareo druggy coming towards the end of his term. So he might not get many more chances to help out in terms of supporting European economies, which have been underperforming. So it's expected to offer some fresh long-term loans at decent rates for people that may help to stimulate things, but really the equity market reaction that would probably determine where we go. So if you're a paean stocks take it. Well, I live, so they can get a bit attraction than probably the, the rally we've seen in European bonds will probably take hold. But if the equity market is a bit more. So fit shrugs the decision than the ready will probably continue. So really how other asset classes perform moti Termine, but certainly, a lot of jitters around because also the Europeans are watching. What's going on, of course, in America, as well and trade talks on various fronts. So there's quite a few moving parts of the moment, right in as the Italians kind of puff up their chests, and, you know, double dare the EU to really do something about budgets. It's, it's really a fascinating game

He dissolved parliament and went on to give a speech about the way forward with a mole on joined by Nina dunk of its who is global fellow at the Wilson center, an expert on disinformation, and the former in the former communist space. She joins us from Vilnius Nina. We know that Celeski chose his spokesperson through social media will know that he's dissolved parliament will they be a snap election in the traditional way or always to expect something entirely different. Well, I'm not even one hundred percent short there will be a snap election, actually, because based on the constitution of Ukraine, given that the parliamentary coalition collapsed last week on Friday, technically the president is not allowed to dissolve parliament, although the preparations for an election in July seem to be going forward. And there was a lot of jubilation in the crowd gathered outside of Ukrainian parliament. Yesterday when this was announced, so we'll see. But certainly this presidency is off to a traditional start. "Another way was the literal destruction of communities. The city of Vilnius in what's now. Lithuania is one case in point. Vilnius was once called the Jerusalem of the north because of the strength and vitality of its Jewish culture and learning and at the heart of Jewish Vilnius was the great synagogue are Schule in Yiddish the Nazis burnt down during World War Two. Then after the war, the Soviets level the ruins and built a school on top of it for three years. Now, archaeologists have been working on the site and the progress has been pretty remarkable. Richard Freund is part of the team Freund as a professor of Jewish history at the university of Hartford in Connecticut. I s m I about the historic significance Vilnius in Jewish culture. It was very dynamic location, and it was not only because of the religious life was because add cultural life. You had Zionists and you had secular Yiddish culture and literature and music. But the thing that was the epitome, the symbol of the story was this great synagogue and Schule Hof, which was a combination between a ninety. Second street y assisting chapel and the Vatican it was the size of a football field. And it was one of the great institutions of Europe in its time. You're the reference to the ninety. Second street y, we should underscore. It's not just a place to sleep like a hassle, but gathering space for all kinds of cultural events. Prior to World War Two, how old was a Jewish community and Vilnius and how old was a great synagogue? The Jewish community at Rives somewhere in the fifteenth century, and the first synagogues were established were were would. But the the final disposition of a great synagogue was probably in the seventeenth century. And so for about four hundred years, you have this institution that sitting in the middle of downtown Vilnius which was across. Roads of Europe and the development of the of the the culture was that not only was that a synagogue also had inside of the synagogue at had twelve smaller, synagogues and at every different cultural institution. And then it had a mass bath house in the back because the Jews and the Jewish quarter didn't have water in their homes. So they had to go at least three times a day to drink, go to the bathroom, take showers, and some people even went there to pray. Yes. So the ninety second street, y metaphor is really apt toes. What kind of progress you in the archaeologists are making? Well, you know, I'm a field archaeologist, that and we have a, the university of Hartford, has a geo physics and science team that goes out to sites. We've now done fifteen sites just in with the wanian forty, five sites worldwide. We go to a site and we can map the subsurface using ground penetrating radar and electrical relatively tomography, and no. No, what's there before we've excavate? And that's really what made this work so possible because the great synagogue was located underneath and elementary school downtown Vilnius. And but for this equipment, we wouldn't have been able to not only prove it, but then we were able to excavate in the play ground all of the artifacts. And once the the government saw just how many artifacts will coming out of this, they decided to close the school. Yeah, it sounds like the technology was crucial to not just as discovery but getting permission to do this research. And I think that this is the the one thing that is very important is the holocaust and genocide sites. When people think that everything is destroyed, actually may still be there below the surface. And this gives us an up between ity to go to the place. See if it's there. If it's burials, we can at least Mark them. So they're not desecrated again, but also you can excavate sites like the great sin. Agog we've already taken out hundreds of thousands of small fines and the great altar. The great Bima that was taken who has found this this summer really is remarkable as if we found Westminster Abbey buried underneath rubble. So the whole thing is that the great synagogue had secret..
