Non-Animal Origin cell culture supplements and manufacturing aids for biologics manufacturing

"Welcome TO THE CELL. Culture dish podcast. Non Animal origin cell culture supplement in manufacturing AIDS for biologics manufacturing. I'm Brandy Sergeant Editor of Cell Culture Dish joining me. Today is Dr Tobias Hurtig Regulatory Affairs Manager and Dr Tillman Product Manager Supplement in manufacturing and both with mark. Kgi Darmstadt Germany. I wanted to start today by asking if you could give us. A definition of non animal origin. Is there an industry wide understanding of the term? I would love to give you a definition for anymore origin or industry-wide definition of for anymore origin. Free or non anymore origin. Unfortunately there is no definition and there is no useful or clear. Definition provided by the regulator switch rate. Cipolla provide you a definition for animal origin. But not for non any minority with that in mind. What are the distinctions between primary secondary and tertiary levels for non an origin? So these determine partners attach level non anymore our Chin or animal origin. Free these terms are used to what people try to distinguish how far away they believe. The actual origin is for instance primary non anymore. Origin would mean okay. Something which is not directly derived from animal secondary on animal origin would be used when you want to say okay. He has something for instance last week. Produce Feminization and dissemination media did not contain any animal orchard material for instance recumbent insulin produced by bacteria. And if I may use the recumbent intimate as an example for tests and that could be used when you want to say okay. He is pro instantly which needs kieft into instantly by an enzyme. And this enzyme that I'm using a knot of animal origin and be also is not coming out of feminization which an arched material in media. These terms are not fully defined. So if you read about these terms you want to make sure that you really understand what the author or the user actually wants to tell you. Thank you for that. That's helpful as a basis for discussion. I was hoping you could also clarify vice for for listeners. Why is animal origin? Such a big concern in the cell culture realm let you start with why Edema or a Chin a concern. Animal origin is concerned. Typically with the from sound are adventitious agents coming with that animal material Dr However other concerns for instance dictate religious concerns halal kosher concerns around the origin. That could be lifestyle concerns that you think but we can waive alleged. Cheese are CO ever. These could also allergies. Ause topic focuses planned to proteins but they major issue are adventitious agents which could be so Nautica so they could cross the species barrier from the animal to human to the patient and causes disease with the patient These competitions agents might be viruses for instance sonars. A rabies are viruses that I've known to cross the species barrier or a protein protein-based agency like There the priors causing TSE pse. Other concerns are related to the supply situation so getting animal origin raw material. Importing that into your country could be associated with restrictions likewise she ever media produced which contains unlimited died might be import-restricted into the country. Where your customers sitting with the got to the Cell Culture? There is a another concern. And that is these the celts. The mammalian cell Kasha might be susceptible to such viruses. And if you OUGHTA robbing a nephew witcher on a bio rector unachieved g conditions. You need to tightly control your by tour and a virus might have a whatsoever effect to yourself. Kasha and from that point on your cell culture would not be regarded as p compliance. We talked a little bit about the fact that there isn't a regulatory definition of non animal origin. But I'm hoping that you could share with us. What the regulatory view on animal origin components is the regulatory few on Non Amel components is of course that this is the preferred As a regulatory person you would like to avoid any more mature whenever possible or you would expect that any moment you to comply with all the Pantin interrelations with regard to non animal materials and Lesser Regulatory Person. I feel that as much as I appreciate it. I always cautious because I don't want to end. Wishful thinking and due diligence is always required. Are those non animal materials. Don't get blind folded around the the documentation truly want to understand why this material is known in origin. So keep your eyes. Open your mind work with your suppliers as close as possible to truly understand when they talking about nine non anymore origin. Is that exactly what you want to half what you think? Non Animal Arjun is to avoid any ambiguity and confusion and at the end natives disappointment. I also feel like Worthwhile when you're auditing your vendors to question the the animal or non Hemel origin situation and to make sure that you're suppliers are in fact producing S. You wish to

"Right. Look, it's it's really difficult. I afford, for example, lots of the known AMP kind activties are on much higher doses of mid for me. Well, that's my point is can you clinically match them. And no because you you really don't know at the end how much mid forming easing the sill what we're doing. Now, we're doing isolated sales. In other words, we treated animals with forming, and we're taking is elated sales to just see the ability of of with formula individuals sales in amongst homogeneous tissues. Yeah. Like, we'll take pot sides sites and look at the very expression. Yeah. Right. So it's all kind of important question. But but the story still I'll tell you the story because at the end I'm going to answer you what he's doing for aging. Okay. And I don't want to rush this story because this is a great story you take your sweet time. So think about it in a scheme. You have the mid forming in the middle getting through the plasma membrane getting to the mitral congre. And on the left side. Let's say there's the Ampy kindness. Okay. Now in the right side of sorry. Just for the listener explain what AMP kind as does the high level. It's it's a hormone of what it's a hormone of nutrient deprivation, and therefore it tells the body to do what like when you don't eat for a day AM PK goes up. Why will? But it's also an exercise me medic pathway. Yes. That's what happens with exercise. Really? That's was the the hope of what the aim behind these doing. But it's. A nutrient sensing that these impart upstream of 'em Tor. Okay. But the other side of mid forming is what happens to the might to country of because in a way. And I hope I won't regret saying it here, but mitt formulas the weakside night. Okay. Look what cyanide acted at complex four. Right. But now, you just mean more broadly speaking heading the right eighteen Heeb it, so yeah, there's less arrests production, okay in then, there's less inflammation. And there's other things that's going just because it might country are his list of see the Tiv pathway going down. So this is a great example of why binary thinking doesn't very doesn't do very well in biology, right? It can't be all or none will. So now, I can go on and on and connect, although sweet due to eventually. A pillar of a, gene. But I wanna insert another thing because other things are happening, for example, insulin levels goes down. Right. Why is it just due to the reduction of paddock Lukasz output? I'll tell you why I think and then also inflammatory factors are going down. And there's a whole NF Cup be action of mid forming that might be part of the hour s and part could be independent even without my congre. Okay. It's confusing. But this is the point I wanna make I don't know which one of those is important for aging, but parts of what you're measuring is the following yueh-fei the aging on a cellular level. Okay. So the younger that the sales are younger a lot of things are correcting themselves. So I think at the end the lower insulin levels low inflammatory are not necessarily direct effect. And that's why we're we're fighting all the time a buck multiform these doing because you can measure lots of things, but the things you're measuring because aging was fixed and once agents fixed, then there's whole Hemel dynamic readjusted, or whatever you wanna call eat. And then you're measuring that everything, and this is not only typical mid form this with rapamycin with veritable. You also can see that. There's a lot of things that are improving themselves. And I think we have to get used to the FEC that when you have a drug that we argue what it's doing because everybody's measuring something some of it is true. But it's like secondary. It's you fixed it richer, and unrelated, and it's funny. You mention those three which again, I think speaks to an advantage you have with tame. So when you look at rapamycin, reservoir, trawl and metformin the big advantage of metformin. Is you already know the dose and the frequency with rapamycin over dinner tonight. I'm gonna give you my philosophy on the dose and the frequency than we can discuss it. But I can't demonstrate it to you with anywhere near the validity that you could do the same with metformin at wonderful discussion with David Sinclair and bioavailability came up on resveratrol. Right..

All right. Okay. What the finger again, you know that. Okay. What kind of bears doesn't have any teeth? About it. Think about it gummy bears. All right, good. They'd gone, kiddo. Thanks. All right. Listen for the oh, we got. We got Ryan back. Yes. Yes. We're going to head back to Ryan litterbox issues. And we're back. Yeah. I'm here. All right. Okay. So now, do you suspect either one of the cats are, you know, one over the other what I'm suspecting the older cat? She's she seems to be more of a kind of a loner type versus the younger one. I think she's not appreciating the company is much. Yeah. Definitely definitely nowadays. Is this a daily occurrence a weekly occurrence what? Yeah. They've been together for about a week. And it's. It's kind of a few days in a row here. All right. But you might want now is a kitten or the two year old is that what always available to the female. I mean, did you separate them at all? Or did. You just pray for the best. Yeah. For the first. Three days we had the female on a separate room by herself. We opened the door and let them kind of have it out. And they seem to be getting along, you know, now after spending some time around each other. But. Yeah. Cats canes, slowly and also true when an older cats been an older cat for a long time. Raolison? Here comes this little son of a gun. Dickens, and that unnerves anybody nerves legged people were having little ones around us. It's like oh Calgon take me away. So what you got to make sure that kitchen does not the two year old doesn't have access to the eleven year old all the time that you give that cat breaks. Alright. So that and sometimes what you're really lucky because usually it takes two to three weeks of like, the one cat stays in the room and the one cat stays in the house. Okay. Then a couple of times a day, you switch places where the older cat would go into the room. There were the younger cat was in and the younger K. So they trading places where they can get you see each other, smells and such like that. Because usually the slower you go the more chance of it working is what happens so they just kind of get used to late you put one in the kennel so one can sniff the other without the other one get wacked. But so basically, that's what it is. Now, have you changed the litter at all? The same litter that you're using. Let's see I guess I guess the one hasn't been completely changed out yet. And that's I guess that's the one they're both using. And then are you using litter that is really perfumey smelly? I guess a little bit of sense because cats and everybody's got cats. Don't buy that stuff. Just get stuff that with no set. And then I get the big bags of baking soda, you know, like, SAM's or whatever and sprinkle a little bit on the bottom and put my. My litter in and then I sprinkle a little bit on top. And then the cats walk it in and and that takes care of any order right now, they're finally coming out with older free that controls older. You know what? I mean, the because most has hate that that's meant. That's smelly. They just think about it be like every time you go. You know, you go to use your toilet. Somebody shoved one of those fragrant things in your nose. Kind of like get it out of there. And so then what food do you feed him? They're each. They're just getting a dry food deputy. We didn't change it at all from what they were doing. So and they seem to be each eating from third their dishes, you know, but it's the coming back out. We're having the issue. Okay. Now, you giving set feeding times free feeding. Is basically always always food out for. Okay. Because what you got to remember is that when you leave food out. It's it gets stale gets rancid, you know, with the with the oxygen. So every cat should have this a set feeding times once in the morning once at night and remember cats, especially seven year old. You know, if he's always been feeding dry matter. That's like a member cats came from the desert, they get the moisture from what they eat by eating dry food with all the carbohydrates over time. They start getting crystals in their urine and start having Marino problems. So you do really well if you'd switch to a gray canned grain, free, diet half a can in the morning and a half a can at night for the older one. And then how is how many pounds is your older one? Probably around thirteen four is she overweight? Probably a tad. Okay. Because there by going to the grain free, canned good food, the cat will lose weight because she's not getting the carbs anymore that turned into sugar and sugar turns into fat right as so. And then like with the young the two year old, my guests are probably not quite a half. A can you know in the morning and a half a cat at night. Did you just have to play with? But the thing is is that the doodoo this left for you to obviously pick up is it is it very dry, or is it what what's. I guess when I I found that it's it's seems dry. But at the same time, I don't know for sure how right right, right, right. Because sometimes if a cat is really having a problem is best to put a litterbox there. And then try to work the litterbox back to where you want. So you don't have because you don't know their PM either, you know, and the anti icky poo was the best stuff to pick that up with all right? That changes a chemical pulp component urine or doodles or any smell what if naturally, but so I guess what I would do is I try to agreed free canned food. I would put a litter box where the the cat seems to be going. And then change that make the to non perfume e and then make sure that the young one doesn't have access to the older one all the time to give the older one a break and not that needle put them into Spanish city because you can't get rid of you'll get outta my face and get off of me. Right. And so we're let's say you're you've got the older one and another room, and you're watching TV and the younger ones somebody else's playing with right or vice versa or Gedo just put the younger one into a room and give the change it up. So that the cat has access to be able to get away. Okay. But anyway, and like I say, my guess is behavior thing there is a book called your cat. By Hodgkin's, H, O D G K I N S and Nesta bible for all cat owners for feeding and for vetting and for behavioral problems. It's called your cat. Okay. So try that. And that's but I like I said sometimes right? You're putting my guess is just is stresses what's causing the not good stuff. So okay. And then also true by going to grain free, canned food, you only in a couple of weeks, you will notice a Colt on the cats will be phenomenal. Where they will still seem like they're not shedding as much. I had eight cat sleep in my house at one time. Nobody knew where the cats were sleeping because all the cats coat was the cats and dogs should only shed twice a year. Was this spring wants to the Paul and they're shedding perpetually? Then like, I say, I dry food with cats. Does it and then they also need some fish oils to okay? So. Yeah. But get that book your cats. All right. Great. Okay. Good luck. Have a great day you too. "Absolutely as the main character of this film he's offscreen quite a long but he drives and if you look at this film i think the three extra ture applies to him and it makes sense that it ends with him because he has jeeves ultimate victory and now he's about to celebrate up i i might i might i might be wrong but it's kind of like i think he doesn't hit all the beats of the hero's journey in this movie could chunk of them good chunk yeah but all right all of them that that this this quite a lot of that journey that we used to the hero taking the uc the villain go through and that's why it was really interesting about this film but going back to the impact of the ending i mean just totally the way kind of this this is the the music stops and then you just going to call this of wind on on the soundtracks behind behind as it's happening you just goes quiet and then he ends quietly you know and then he the school comes out in the end credits by seeing it with the audience at the senior bowl hemel hempstead you know it was just like the room stayed quiet through those credits the impact was less than slightly when this thing happened and then someone shouted superman which point i really wanted to become the complete dick no i think you'll find a different universe there it is of course captain marvel captain marvel as opposed to mar vel and the design of her uniform as done of course by jamie mckelvie who is very excited to see his design even on a pager which is very cool he go to page as well going your bunk balances.
